HiSeq 2000 Illumina
The Hiseq 2000 is the new next generation sequencer from Illumina to follow up the GAII. It can now hold two flowcell in one machine. Each flowcell can be scanned on two sides, increasing the amount of output per lane. With a 2x 100 bp reads the Hiseq can produce up to 200 Gb in ~8 days. The flowcells can operate independently from each other with different protocols and with separate reagents.
Ion Torrent PGM ("Personal Genome Machine")
The PGM is a desktop sequence machine that uses disruptive semiconductors sensors to give a real-time measurement of hydrogen ions that are released during DNA template extension. A high-density array of wells on the Ion semiconductor chips provides millions of individual reactors while an integrated fluidics system allows reagents to flow over the sensor array. The direct measurement of hydrogen ions circumvents the need for fluorescent labeled nucleotides and time consuming imaging, thus reducing sequencing time.
For now the PGM is able to sequence ~100 bp DNA fragments (or amplicons) in less than 3 hours with an output of 10 Mb per 314 chip. This will quickly improve over time to longer fragments up to 400 bp and a 1 Gb output.
